Buy ALDH1A2 antibody involved in normal retinoid metabolism have been shown to be reduced in prostate cancer, suggesting that changes in the regulation of these pathways are associated with tumorigenesis. Treatment of the TRAMP-derived C2N cell line with retinoic acid has been shown to increase cell differentiation and decrease growth in vitro, and in vivo it has been shown to reduce tumor progression in the prostate in the TRAMP model. Therefore, we wished to examine the expression of several of these genes and their relationship to carcinogenesis in the TRAMP model.

Quantitative RT-PCR was used to measure the levels of ALDH1A1, ALDH1A2, and ALDH1A3 mRNAs in prostate lobes from three nontransgenic age-matched littermate control mice (WT) and three TRAMP transgenic positive mice at 18, 24, and 36 weeks of age. The expression levels of these genes were compared to mRNA for the ribosomal phosphoprotein 36B4 as an internal control.

Immunohistochemical staining showed that the ALDH1A2 protein is primarily localized in the cytoplasm of normal prostate epithelial cells. In contrast, the ALDH1A2 protein was weakly detected in the cytoplasm of prostate cancer cells in the DP, LP, and AP lobes of TRAMP mice, with a loss of nuclear localization (Figure 2A).

The ALDH1A2 protein was also detected by Western blot analysis of paraffin-embedded human prostate tissue. There was little to no ALDH1A2 protein detectable in the malignant tissues compared to adjacent benign tissue (Figure 6A-F). Interestingly, there were two different isoforms of the human ALDH1A2 gene. The full length gene product, and the N-terminally truncated isoform (also known as RALDH2-T) have identical mRNA sequences, suggesting that they may share some functional characteristics.
